Forensic Applications of Whole Genome Amplification

چکیده

Whole genome amplification (WGA) has been touted as a possible technique to augment fragment analysis of STRs in amplifying low-levels of DNA recovered from crime scenes or items. Only LCN DNA analysis is routinely used as an additional amplification step; but this is not without problems, such as the increased incidents of stochastic variations. A review of the literature was carried out in order to assess the latest research and to identify a potentially fit for purpose whole genome amplification technique. There are three potential alternative techniques which show promise, miPEP, dcDOP-PCR and MDA used in conjunction with a macromolecular crowder; with the majority of studies investigating the effect of non-crowded MDA on a variety of stains, including degraded, low-level and stains containing

Random Whole Metagenomic Sequencing for Forensic Discrimination of Soils

Here we assess the ability of random whole metagenomic sequencing approaches to discriminate between similar soils from two geographically distinct urban sites for application in forensic science. Repeat samples from two parklands in residential areas separated by approximately 3 km were collected and the DNA was extracted. Shotgun, whole genome amplification (WGA) and single arbitrarily primed...

Digital Droplet Multiple Displacement Amplification (ddMDA) for Whole Genome Sequencing of Limited DNA Samples

Multiple displacement amplification (MDA) is a widely used technique for amplification of DNA from samples containing limited amounts of DNA (e.g., uncultivable microbes or clinical samples) before whole genome sequencing. Despite its advantages of high yield and fidelity, it suffers from high amplification bias and non-specific amplification when amplifying sub-nanogram of template DNA. Here, ...

Evaluation of circular DNA substrates for whole genome amplification prior to forensic analysis.

Forensic biological evidence often contains low quantities of DNA or substantially degraded DNA which makes samples refractory to genotype analysis. One approach that shows promise to overcome the limited quantity of DNA is whole genome amplification (WGA). One WGA technique, termed rolling circle amplification (RCA), involves the amplification of circular DNA fragments and this study evaluates...

Applications of multiplex ligation-dependent probe amplification (MLPA) method in diagnosis of cancer and genetic disorders

Introduction: Lots of human diseases and syndromes result from partial or complete gene deletions and duplications or changes of certain specific chromosomal sequences. Many various methods are used to study the chromosomal aberrations including Comparative Genomic Hybridization (CGH), Fluorescent in Situ Hybridization (FISH), Southern blots, Multiplex Amplifiable Probe Hybridisation (MAP...

A graphical simulation model of the entire DNA process associated with the analysis of short tandem repeat loci

The use of expert systems to interpret short tandem repeat DNA profiles in forensic, medical and ancient DNA applications is becoming increasingly prevalent as high-throughput analytical systems generate large amounts of data that are time-consuming to process. With special reference to low copy number (LCN) applications, we use a graphical model to simulate stochastic variation associated with...
